Cabenuva is an injectable medication that is administered by a healthcare provider. It is FDA-approved as a complete regimen for the treatment of HIV-1 infection in adults and adolescents 12 years and older who weigh at least 77 pounds to replace the current medication regimen in appropriate situations as determined by the doctor.
This medication kit contains two medications: cabotegravir, which is an HIV integrase inhibitor, and rilpivirine, which is a non-nucleoside reverse transcriptase inhibitor, or NNRTI. The two medications work together to decrease the amount of HIV in the blood. They do not cure HIV but may decrease your chance of developing acquired immunodeficiency syndrome (AIDS) and HIV-related conditions like serious infections or cancer. These medications are used along with lifestyle changes and safer sex.
Before starting Cabenuva, you may take daily pills by mouth for one month to see how your body reacts. Cabenuva is administered by your healthcare provider as two injections, one injection into the muscle of each side of your buttocks. Depending on the treatment plan, you will receive an injection either every month—or every other month (after the first two injections, which are one month apart). Cabenuva is formulated as a long-acting medication that helps you stay undetectable (where the amount of HIV in the blood cannot be measured by a lab test) and not have to take daily medication. To stay undetectable, you must keep your appointments and receive your injections on schedule. You will keep receiving Cabenuva injections unless your healthcare provider advises another type of treatment. If you stop taking Cabenuva, you will have to take other medications to treat HIV-1 infection and lower the chance of developing resistance.
Common side effects of Cabenuva include:
Injection site reactions like pain, swelling, and redness
Fever
Fatigue
Headache
Pain in the bones, joints, ligaments, tendons, or muscles
Nausea
Trouble sleeping
Dizziness
Rash
After giving you an injection, your healthcare provider will monitor you for at least ten minutes in case you have a reaction. Symptoms of a post-injection reaction may include:
Difficulty breathing
Anxiety
A warm feeling
Rash
Sweating
Stomach cramps
Back and chest pain
Feeling numb in or around the mouth
Lightheadedness
High or low blood pressure
If you have symptoms of an allergic reaction at any time while you are on Cabenuva, get emergency medical help. These symptoms may include hives, difficulty breathing or swallowing, or swelling of the face, lips, tongue, or throat.
Other serious side effects may include liver problems or suicidal thoughts and behaviors. Be alert to any changes in mood and behavior, and notify your healthcare provider right away if you notice any changes.

A small amount of weight gain is a known side effect of Cabenuva. In clinical studies, people who took Cabenuva gained about 2 or 3 pounds.
You should not take Cabenuva if you are allergic to either cabotegravir or rilpivirine, or any ingredient in Cabenuva. You should not take Cabenuva if you take any of the following medications, which are associated with drug interactions that would make Cabenuva not work properly:
The anticonvulsants Tegretol (carbamazepine), Trileptal (oxcarbazepine), phenobarbital, or Dilantain (phenytoin)
Rifampin
Steroids such as dexamethasone
The dietary supplement St. John’s wort
There are other drug interactions that may occur with Cabenuva, so before you take it, make sure your healthcare provider knows about all the medications you take, including prescription and OTC drugs, vitamins, and supplements.
Cabenuva can be used in people who are 12 years and older and weigh at least 77 pounds to replace their current HIV-1 medications when the healthcare provider determines the treatment is appropriate. Patients must:
Be virologically suppressed (with an HIV-1 RNA of less than 50 copies per mL)
Be on a stable antiretroviral medication regimen
Not have a history of treatment failure
Not be resistant to cabotegravir or rilpivirine
