Lupron Depot Ped (leuprolide acetate) is a brand-name prescription drug prescribed to block puberty in children diagnosed with Central Precocious Puberty (CPP). Children are diagnosed with CPP if sexual characteristics begin to develop too early (before the age of 8 in girls and 9 in boys), blood tests show higher levels of luteinizing hormone (the hormone that stimulates the production of sex hormones), and tests confirm the absence of other possible causes, such as tumors.
Leuprolide acetate, the active ingredient in Lupron Depot Ped, blocks the production of male and female sex hormones. The process starts in the hypothalamus, a structure in the brain that secretes a hormone called gonadotropin-releasing hormone (GnRH). This hormone attaches to cells on the pituitary gland which then produces luteinizing hormone (LH). Luteinizing hormone in turn “switches on” the tissues that produce male or female sex hormones. Leuprolide attaches to cells on the pituitary gland that GnRH would attach to, so the pituitary gland produces less luteinizing hormone it produces. The amount of sex hormones in the body falls and secondary sex characteristics stop developing.
Lupron Depot Ped is also used off-label as a puberty-blocker to prevent normal-onset puberty in children with gender dysphoria.
