Repeated or recurrent seizures are the basic criteria for a diagnosis of epilepsy, a nervous system disorder. Seizures are caused by the repetitive, overexcited, and abnormal firing of nerve cells in the brain, usually triggered by a situation or other stimulus. The active ingredient in Sabril, vigabatrin, works by increasing a chemical in the brain that prevents signals from passing from one nerve to another.
Because of the potential for severe and debilitating side effects, patients cannot fill a Sabril prescription until they have signed a patient-physician agreement as part of the Vigabatrin REMS program. The prescription is sent only to specialty pharmacies certified by this program. The prescription is sent by overnight courier to the patient.

The most common side effects of Sabril include headache, dizziness, drowsiness, feeling tired, fatigue, infections, swelling, loss of coordination, joint pain, nausea, vomiting, weight gain, diarrhea, problems sleeping, tremors, blurred vision, double vision, flu symptoms, and cough.
Mental and mood changes are common side effects and can be severe. The most common are memory problems, depression, irritability, confusion, abnormal thoughts, abnormal behavior, abnormal dreams, disorientation, and problems using language. Less common but more severe mental and mood changes include psychosis (hallucinations and delusions), delirium, and low-level mania.
Serious side effects of Sabril include anemia, nerve damage, hearing loss, and vision loss. Although rare, Sabril can cause permanent and sometimes debilitating vision loss (tunnel vision and blurred vision). This can happen unpredictably and, while the risk increases with dose and length of treatment, vision loss can happen at any dose for any length of treatment, including a single dose. Vision loss is usually not detected until it is severe. Once detected, vision loss is not reversible. Vision tests will be given at the start of Sabril therapy and will be repeated every 3 months. However, you must immediately inform your prescribing physician if you notice any changes in your vision while on Sabril.
Allergic reactions are rare but include hives, rashes, itching, and trouble breathing. Severe allergic reactions are possible, including toxic epidermal necrolysis and Stevens-Johnson syndrome, potentially hazardous drug reactions that cause dark rash, blistering, peeling, and skin death throughout the body as well as fever, sore throat, and flu-like symptoms.
This is not a complete list of side effects. Because of potentially severe side effects, your prescribing physician is required by the Vigabatrin REMS program to thoroughly explain the side effects and how you should manage them. Your physician will also give you a printed guide to vigabatrin treatment. You will be required to fill out an agreement with your physician that certifies you understand the side effects of Sabril. Make sure all your questions are answered before you sign the patient-physician agreement.
